Blair loses her virginity to Chuck Bass, at the end of the seventh episode of season one, "Victor, Victrola". She breaks up with Nate earlier in the episode. The majority of the first season focusses on the Nate/Blair/Chuck Amore triangle, and, in my opinion, it's the best period of the show.

Ya I heard season 1 was really good as I just started it, but is true Amore between Blair & Chuck??
Season One is the best, Season Two is not bad, Season Three is quite terrible. Yes, it is "true love" between Chuck and Blair. They become the show's main couple, and have storylines featuring eachother and their relationship throughout all the seasons.
